Frequently Asked Questions Regarding our mRNA Enzymes

  1. What products do you offer for an IVT reaction? 
    1. We have Restriction Enzymes (BsaI, BspQI, Kpnl, Nrul, Pvull, Sall, Sapl, Xbal) for plasmid linearization and IVT enzymes including T7 RNA Polymerase, Murine RNase Inhibitor, Pyrophosphatase, DNase I, and a capping system that includes mRNA Cap 2’-O-Methyltransferase and Vaccinia capping enzyme.
  2.         What capping system is suitable for your IVT reaction? 
    1. KACTUS T7 RNA polymerase supports both a cotranscriptional using cap analog and vaccinia capping system. We have two GMP grade capping enzymes: mRNA Cap 2’o-Methyltransferase and Vaccinia capping enzyme.
  3.         Does KACTUS T7 RNA Polymerase support different forms of RNA?
    1. Yes! KACTUS T7 RNA Polymerase can be used for linear mRNA, circular RNA, self amplifying RNA, and more.
  4.         What GMP grade products for IVT does KACTUS have?
    1. KACTUS has GMP-grade T7 RNA Polymerase, Murine RNase Inhibitor, Pyrophosphatase, DNase I. 
  5.         Does KACTUS have T7 RNA Polymerase variants? 
    1. Yes, we are currently trying to find variants that have high yield and less double-stranded RNA. Currently we have 4 variants that have different applications. We have a variant optimized for circRNA, and multiple variants that work well with long RNAs. Please contact us at sales@kactusbio.us to learn more about getting a sample to test!
  6.         How should the IVT enzymes be stored?
    1. The IVT enzymes should all be stored in a -20C freezer for 24 months. 
  7.         There is an enzyme I want that is not GMP-grade. Can you make this GMP grade? 
    1. Yes, we can scale up a GMP grade enzyme based on the enzyme and quantity needed. Please submit a custom protein request form
  8.         What is the difference between GMP-ready and GMP-grade?
    1. GMP-Grade: Manufactured according to cGMP guidelines.
    2. GMP-Ready: Industrial-grade manufacturing transferrable to GMP-level manufacturing.
  9.         Does KACTUS provide a buffer with the T7 RNA Polymerase?
    1. Yes, the buffer provided with the GMP T7 RNA Polymerase was highly optimized and components are confidential.
  10.       What is the concentration of the T7 RNA Polymerase as well as the variants? 
    1. The concentration of all T7 RNA polymerase (including our variants) is 50U/uL.
  11.       Do you provide a recommended protocol for using your IVT reagents? 
    1. Yes, this can be found on the product page where there is an option to download the “Datasheet".
